For example, about 99% of CML leukemia patients have the Philadelphia chromosome in their bone marrow cells, and the translocation 15;17 abnormality is found only in the APL-M3 subtype of leukemia. It can also report the percentage of cells with normal and abnormal chromosomes. Chromosome analysis therefore helps classify the disease type and prognosis, which is useful for physicians' treatment decisions.

Test limitations

Chromosome diagnosis or analysis may not meet the defined standard because of the patient's own condition, for example, patient on medication, patient expired at collection, etc., which may result in no metaphase or a low mitotic index.

Sample collection method

Method 1

1. Use a sterile disposable 5 cc syringe; draw up heparin to rinse the syringe thoroughly, then squeeze out all excess heparin, leaving only a small amount coating the syringe. Change to a new needle and prepare to draw the patient's blood or bone marrow.

2. (For blood) Wash the arm to be drawn clean with soap and wipe with Betadine, using sterile precautions as for a blood culture. If Betadine is unavailable, alcohol may be used, but let it dry before drawing blood.

3. Draw blood / aspirate about 3-5 cc of bone marrow into the syringe, cap the needle tightly, shake to mix the blood/marrow with the heparin, and send the whole syringe for testing.

Method 2

Draw 3-5 cc of blood into a sterile tube containing a sodium heparin or lithium heparin anticoagulant.

Every time a sample is submitted for testing, the details on the test request form must be specified as clearly and completely as possible.
